Darstellung und Eigenschaften der Coenzymanalogen [4-(3-Acetylpyridinio)butyl]-adenosin-pyrophosphat und [4-(3-Bromacetylpyridinio)butyl]-adenosin-pyrophosphat

摘   要:

1-(4-Acetoxy-n-butyl)-3-acetylpyridiniumchlorid (1) wird aus 3-Acetylpyridin und 4-Chlor-1-butyl-acetat hergestellt. Nach Hydrolyse der Estergruppe entsteht durch Behandeln mit Metaphosphorsure das Phosphat 3, das mit Adenosinmonophosphat zum Coenzymmodell 4 kondensiert wird. Mit Dehydrogenasen bilden 4 und Dihydro-4 stark dissoziierte Komplexe, deren Fluoreszenzspektren Verminderung der Protein- und Steigerung der Acetyldihydropyridin-Fluoreszenz zeigen. Im Test mit Alkohol-Dehydrogenasen aus Hefe und Leber sowie Lactat- und Glycerinaldehyd-3-phosphat-Dehydrogenase1) sind diese Verbindungen als Coenzyme schwach wirksam. Mit Brom reagiert 4 unter Bildung des Bromacetylderivats 5, das die Alkohol-Dehydrogenasen aus Leber und Hefe inaktiviert und mit Glycerinaldehyd-3-phosphat-Dehydrogenase stabile (Coenzym-Enzym)-Verbindungen bildet. Durch Behandeln mit Dithionit wird das eingebaute Coenzymanaloge in die Dihydroform übergeführt. Die optischen Eigenschaften der (Dihydrocoenzym-Enzym)-Verbindungen entsprechen denen der reversiblen [(Dihydro-4)-Enzym]-Komplexe.

Synthesis and Properties of the Coenzyme Analogues [4-(3-Acetylpyridinio)butyl]adenosine Pyrophosphate and [4-(3-Bromoacetylpyridinio)butyl]adenosine Pyrophosphate

1-(4-Acetoxy-n-butyl)-3-acetylpyridinium chloride (1) is prepared by condensation of 3-acetyl-pyridine with 4-chloro-1-butyl acetate; the acetyl group is removed by acid hydrolysis. Phosphorylation of 2 with metaphosphoric acid is followed by condensation of the derivative 3 with adenosine monophosphate, forming the coenzyme analogue 4. Both 4 and dihydro-4 form strongly dissociated binary complexes with dehydrogenases whose fluorescence spectra show quenching of protein fluorescence, and enhancement of dihydrocoenzyme fluorescence. The analogue 4 and dihydro-4 show little coenzymatic activity in tests with alcohol dehydrogenases from yeast or horse liver, lactate dehydrogenase and glyceraldehyde-3-phosphate dehydrogenase1). Bromination of the acetyl group of 4 results in 5, which inactivates alcohol dehydrogenase from liver and yeast and glyceraldehyde-3-phosphate dehydrogenase and forms covalent coenzyme-enzyme compounds. Treatment of these with sodium dithionite produces the enzyme dihydrocoenzyme compounds having optical properties similar to the binary dihydro-4 enzyme complexes.
