The sterols in three sterol fractions, isolated from rat renal inner medulla after incubation with the labelled precursors [2-14C]acetate or [2-14C]mevalonate, have been identified. The identification has been performed by mass spectrometry in combination with hydrogenation in acid and neutral medium. Additional evidence has been obtained by argentation column chromatography and radio gas-liquid chromatography, and further by ozonolysis for 5α-cholesta-7,24-dien-3β-ol. In conclusion, the inner medulla produces and accumulates in vitro desmosterol (5,24-cholestadien-3β-ol), lathosterol (5α-cholest-7-en-3β-ol) and 5α-cholesta-7,24-dien-3β-ol. In fresh preparations of 1 g rat renal medullary tissue the same three sterols were found in amounts of 10, 3 and 1.4 μg, respectively.
